Overview

ISO/FDIS 25660: Traditional Chinese medicine - Coix lacryma-jobi var. ma-yuen seed is an international standard developed by the International Organization for Standardization (ISO) under technical committee ISO/TC 249, Traditional Chinese medicine. This document establishes minimum quality requirements and test methods for Coix lacryma-jobi var. ma-yuen seed (commonly referred to as Yiyiren), which is widely used as a raw material in traditional Chinese medicine and traded globally as a natural medicinal product.

The standard is designed to improve product quality, ensure safety, and support the effective clinical use and international trade of Coix lacryma-jobi var. ma-yuen seed. It covers the product sold and used primarily as Chinese materia medica and in similar applications where natural medicines are regulated.

Key Topics

ISO/FDIS 25660 covers several critical quality control aspects, including:

  • General Quality Requirements: Seeds should be clean, dry, peeled, and free from visible impurities or contaminants.
  • Macroscopic Characteristics: Clear requirements for size, shape, and exterior appearance to ensure authenticity and consistency.
  • Impurities: The standard sets acceptable limits for non-product matter.
  • Moisture Content: Defines a maximum allowed moisture content to prevent spoilage and degradation.
  • Total Ash: Limits are set for total ash content, ensuring product purity.
  • Identification Techniques: Specifies thin-layer chromatogram (TLC) methods to confirm product identity.
  • Extraction and Marker Compounds: Methods to determine ethanol-soluble extractives and presence of characteristic marker compounds such as triolein.
  • Safety Assessment: Guidelines for testing aflatoxins, pesticide residues, heavy metals (including arsenic, mercury, lead, and cadmium), and sulfur dioxide residues.
  • Packaging and Storage: Requirements for packaging integrity, storage conditions, and the prevention of contamination during transportation.
  • Marking and Labelling: Comprehensive labelling for traceability including product name, origin, batch number, production date, expiry date, and storage instructions.

Applications

The practical value of ISO/FDIS 25660 lies in its ability to harmonize quality and safety requirements for Coix lacryma-jobi var. ma-yuen seeds in international trade. Main applications include:

  • Quality Assurance for Suppliers: Enables producers, processors, and exporters to meet an internationally recognized benchmark for product quality and safety, supporting access to global markets.
  • Regulatory Compliance: Supports regulators in importing and exporting markets to assess product conformity and safety.
  • Pharmaceutical Manufacturing: Assures pharmaceutical and traditional medicine companies of consistent raw material quality, enhancing product safety and efficacy.
  • Research & Development: Provides standardized methodologies for researchers analyzing Coix lacryma-jobi var. ma-yuen seed composition and pharmacological properties.
  • End-User Transparency: Facilitates accurate product labelling and traceability for consumers and healthcare professionals.

Related Standards

ISO/FDIS 25660 is part of a family of standards developed to promote safe and high-quality traditional Chinese medicine products. Related standards include:

  • ISO 23723: General requirements for herbal raw material and materia medica.
  • ISO 1575: Tea - Determination of total ash.
  • ISO 18664: Determination of heavy metals in herbal medicines used in traditional Chinese medicine.
  • ISO 22217: Storage requirements for raw materials and decoction pieces.
  • ISO 22258: Determination of pesticide residues in natural products by gas chromatography.
  • ISO 22283: Determination of aflatoxins in natural products by LC-FLD.
  • ISO 22590: Determination of sulfur dioxide in natural products by titration.

Adoption of ISO/FDIS 25660 ensures consistency across the global supply chain, enhances consumer safety, and supports the reputable use of Coix lacryma-jobi var. ma-yuen seed in traditional medicine.

Introduction
Coix lacryma-jobi var. ma-yuen seed, is the dried ripe seed of Coix lacryma-jobi L. var. ma-yuen (Roman.)
Stapf and is a well-known traditional Chinese medicine. Its Chinese phonetic name is Yiyiren. Currently, it
is commonly used as one of the main traditional Chinese medicines (TCMs) in over 100 prescriptions based
on its wide spectrum of biological and pharmacological activities, for treating edema, athlete's foot, poor
urination, cancer, spleen deficiency and diarrhea, etc. Coix lacryma-jobi var. ma-yuen seed oil extracted
has also been developed as an anti-tumor agent. The global import and export trade volume is huge with
sustainable growth in recent years, and the countries of trade mainly include Republic of Korea, Japan,
United Kingdom, United States, Australia, Thailand, Viet Nam, etc. However, the following concerns exist
regarding the quality and safety control of Coix lacryma-jobi var mayuen seed:
a) The processing method, storage pattern and operation technology of Coix lacryma-jobi var. ma-yuen seed
have not been standardized, which can affect the quality and safety of medicinal materials.
b) The requirements of Coix lacryma-jobi var. ma-yuen seed are different in different countries and regions,
including China, Japan, Korea, America and Europe. The applicability of standards is limited and affects
international trade.
c) Coix lacryma-jobi var. ma-yuen seed ranked the 136th in the priority list of single herbal medicines for
[1]
developing standards in ISO/TR 23975:2025 .
Therefore, an International Standard is needed for the quality control of Coix lacryma-jobi var. ma-yuen seed
and its products.
This document aims to build a systematic and practical international standard for Coix lacryma-jobi var.
ma-yuen seed to control and guarantee stable quality, to ensure the safety and effective use in clinic, to
standardize its products in global trade, and to reduce cases of mycotoxins pollution.
As national implementation can differ, national standards bodies are invited to modify the requirements
given in 5.4 and 5.5 in their national standards. Examples of national and regional values are given in
Annex D.
